IDE 2 is a cell-permeable inducer of definitive endoderm differentiation that induces the differentiation of mouse and human embryonic stem cells into definitive endoderm. IDE2 activates the TGF‑β/Nodal signaling pathway, induces Smad2 phosphorylation, and increases Sox17 and FoxA2 expression. Endoderm induced by IDE 2 integrates into the developing gut tube, expresses gut tube markers, and differentiates into pancreatic progenitor cells. IDE 2 can be used for research related to directed endoderm differentiation of embryonic stem cells.

IDE 2 can induce definitive endoderm from Sox17-dsRed mouse ESCs with an EC50 of 223 nM[1].
IDE 2 (200 nM) directed HUES4 and HUES8 human ESCs toward Sox17+ endoderm, with 57% of cells positive, of which at least 90% co-expressed FoxA2[1].
IDE 2‑induced endoderm (12‑day total differentiation; sequential Indolactam V treatment) differentiates into Pdx1‑positive pancreatic progenitor cells with approximately 51‑53% efficiency[1].
IDE 2 (4-10 days) combined with Nodal enhances Sox17 expression in mouse ESCs, whereas Wnt3a does not[1].
IDE 2 activates the TGF-β/Smad2 signaling pathway in mouse ESCs and upregulates Nodal expression[1].

Kinase activity assays measure the ability of kinases to transfer phosphate groups from ATP to specific substrates, while phosphorylation assays detect the presence and levels of phosphorylated proteins. Phosphorylation can be detected by phospho-specific Western blot, immunoprecipitation kinase assay, phospho-immunofluorescence, phospho-flow cytometry, luminescent ADP detection, radiolabeled ATP incorporation, or reporter-based pathway assays, and these readouts can be applied to cancer cells, primary neurons, mouse tumors, organoids, inflammatory macrophages, ferroptosis studies, and mitophagy studies when the kinase target is biologically relevant.
