CH-123
CH-123 is an orally active lipid-lowering agent. CH-123 inhibits the elevated activities of β-glucuronidase, β-galactosidase, N-acetyl-β-glucosaminidase and acid phosphatase in aortic smooth muscle cells and hepatocytes. CH-123 reduces serum total lipid and cholesterol levels, as well as intracellular cholesterol content in aortic smooth muscle cells. CH-123 significantly inhibits lysosomal enzyme activity. CH-123 can be used in the research of atherosclerosis.

-
Animal Model:unspecified strain (5 animals per group; atherosclerosis induced by 2% cholesterol diet for 4 weeks)[1]
-
Dosage:50 mg/kg
-
Administration:p.o.; daily; 28 days
-
Result:Reduced serum total lipids by ~30% (from 2452.72 mg/100 mL to 1715.90 mg/100 mL).
Reduced serum cholesterol by ~29% (from 801.75 mg/100 mL to 572.27 mg/100 mL).
Left serum triglycerides near cholesterol-fed levels (147.72 mg/100 mL vs. 150.52 mg/100 mL).
Reduced aortic smooth muscle cell cholesterol content by 28% (from 570 μg/mg protein to 412 μg/mg protein).
Restored aortic β-glucuronidase activity to 1.41 munit/mg protein (181% of control), β-galactosidase activity to 1.73 munit/mg protein (244% of control), N-acetyl-β-glucosaminidase activity to 1.39 munit/mg protein (151% of control), and acid phosphatase activity to 11.19 munit/mg protein (189% of control), with no significant difference from control levels.
Restored liver β-glucuronidase activity to 21.17 munit/mg protein (168% of control), β-galactosidase activity to 1.91 munit/mg protein (108% of control), N-acetyl-β-glucosaminidase activity to 1.48 munit/mg protein (140% of control), and acid phosphatase activity to 15.54 munit/mg protein (120% of control), with no significant difference from control levels.
Reduced β-glucuronidase activity in the low-density (1.09 g/cm3) lysosomal fraction of aorta, which was selectively elevated in untreated atherosclerotic rabbits.
