SPRi High Throughput Screening
Surface Plasmon Resonance Imaging (SPRi) high-throughput screening is a label-free, real-time parallel optical sensing technique for biomolecular interaction analysis. It upgrades the single-point detection of conventional SPR to microarray imaging, enabling simultaneous analysis of hundreds to thousands of molecular interactions in a single run, which drastically boosts the efficiency of drug screening and molecular mechanism research. This technology is ideally suited for early-stage large-scale drug screening and fragment screening. It effectively shortens R&D cycles, cuts research costs, and improves the quality of candidate compounds.
Technical Advantages
Experimental Workflow
-
1Preparation of microarray chip: Select compatible chip and densely print thousands of small-molecule compounds or samples on the solid surface to prepare microarrays.
-
2Sample Injection: Inject target proteins into the flow cell. Analytes flow across the chip surface and bind immobilized small molecules for real-time tracking of binding events.
-
3Data Acquisition: Real-time tracking of SPR signal shifts. Full association-dissociation profiles are recorded to produce sensorgrams per array spot.
-
4Data Analysis: Fit sensorgrams from each spot to calculate binding kinetic parameters. Rank compounds by affinity to filter potent hits against target proteins.
