YycG-IN-1
YycG-IN-1 is a selective YycG inhibitor, with an IC50 of 42.9 μM and a Kd of 86.4 μM against YycG from Enterococcus faecalis. YycG-IN-1 inhibits the kinase activity and autophosphorylation activity of YycG, suppresses biofilm formation of Enterococcus faecalis, inhibits the planktonic growth of Enterococcus faecalis, and exerts effects on multiple mutant strains of Enterococcus faecalis. YycG-IN-1 can be used in studies related to Enterococcus faecalis infection.

In Vitro
YycG-IN-1 (compound 16) inhibits the planktonic growth of multiple Gram-positive bacterial strains (Staphylococcus aureus, Staphylococcus epidermidis, Enterococcus faecium, Streptococcus agalactiae) with MIC values of 12.5-25 μM, and exhibits no inhibitory effect on Escherichia coli ATCC 29522 even at concentrations as high as 200 μM[1].
YycG-IN-1 (25 μM; up to 24 h), at its MIC of 25 μM, in combination with Ampicillin (HY-B0522), Vancomycin (HY-B0671), or Linezolid (HY-10394) at concentrations of 1/2 of their respective MICs, rapidly reduces the viable count of planktonic Enterococcus faecalis OG1RF by >3 log10 CFUs within 3 h, and this bactericidal activity persists for more than 24 h[1].
YycG-IN-1 inhibits the formation of mature biofilms of Enterococcus faecalis FB-1 at a concentration of 50 μM, with an inhibition rate of 63.7-85.1%; it significantly inhibits the dynamic biofilm formation of Enterococcus faecalis FB-1 in a fluid shear system at a concentration of 25 μM[1].
YycG-IN-1 reduces the survival rate of biofilm-embedded Enterococcus faecalis cells to 18.29% at a concentration of 50 μM [1].
YycG-IN-1 inhibits the planktonic growth of 257 clinical Enterococcus faecalis strains (including vancomycin-resistant and linezolid-resistant strains), with MIC values of 25-100 μM and MIC50/MIC90 of 50/100 μM[1].
YycG-IN-1 (1/2 × MIC; 24 h) inhibits biofilm formation of 10 strongly biofilm-positive clinical Enterococcus faecalis strains by 74.8%-97.4%[1].
YycG-IN-1 inhibits the kinase activity of purified recombinant YycG′ protein derived from Enterococcus faecalis OG1RF with an IC50 of 42.9 μM; it binds to the purified recombinant YycG′ protein from Enterococcus faecalis OG1RF with a KD value of 86.4 μM[1].
YycG-IN-1 (25 μM; 6 h) downregulates the expression of yycG, yycF and multiple biofilm-associated genes (epbA, epaOX, fsrB, gelE, sprE) in Enterococcus faecalis[1].
