Description

Ac-CoA Synthase-IN-2 is an Ac-CoA Synthase (ACS) inhibitor and antifungal agent. Ac-CoA Synthase-IN-2 binds in the ATP/acetyl-AMP pocket of fungal and human ACS enzymes to exert competitive inhibition with ATP, and inhibits Cryptococcus neoformans CnKbc1-mediated acetoacetate-to-aceto-acetyl CoA conversion. Ac-CoA Synthase-IN-2 can be used for the research of fungal infections[1].

In Vitro

Ac-CoA Synthase-IN-2 (Compound 13) (25-50 μM; variable incubation times) potently inhibits multiple fungal acetyl-CoA synthetases, human ACSS2, and Cryptococcus neoformans CnKbc1, with the highest potency against Coccidioides immitis CiAcs1 (IC50 = 11.9 μM) and CnKbc1 (IC50 = 8.2 μM)[1].
Ac-CoA Synthase-IN-2 (24-72 h) exhibits modest antifungal activity only against Cryptococcus neoformans H99 (MIC = 32 μg/mL)[1].
Ac-CoA Synthase-IN-2 (24 h) shows weak toxicity against HepG2 cells, failing to induce maximal LDH release even at the highest tested concentration[1].
