Adenosine 5'-O-(3-thiotriphosphate)
Adenosine 5'-O-(3-thiotriphosphate) (ATPγS) is an ATP analog. Adenosine 5'-O-(3-thiotriphosphate) can serve as a phosphate donor for Abl tyrosine kinase and ERK2.

In Vitro
ATPγS (0.2-2 mM; 0-180 min) utilization efficiency is much less than that of ATP in the Abl tyrosine kinase reaction with Abltide-GST, with a steady-state thiophosphorylation ratio of about 14%[1].
Adenosine 5'-O-(3-thiotriphosphate) (0.2 mM; 0-60 min) shows differences in utilization efficiency compared to ATP in the ERK2 reaction with MBP, with a thiophosphorylated MBP ratio of approximately 35% at 60 min[1].
ATPγS enables Phos-tag affinity electrophoresis detection of GST-cJun thiophosphorylation by JNK1, demonstrating its capability to capture the thiophosphate moiety of a serine residue[1].
ATPγS (1 mM; 0-240 min; 1-3 days) is accepted as readily as ATP by PhoR in the autophosphorylation reaction, with a steady-state thiophosphorylation ratio of about 48%[1].
ATPγS (1 μM-10 mM; 3 min preincubation and 20 min reaction) inhibits purified C1·C2·forskolin adenylyl cyclase with a Ki of 170 μM in the presence of Mn2+ and 1900 μM in the presence of Mg2+, demonstrating only little affinity gain over ATP under Mn2+ conditions and a substantial loss of affinity under Mg2+ conditions[2].
