AG2 is a glucose uptake tracer and two-photon fluorescent probe. AG2 is taken up by cells via glucose-specific transport systems, rather than passive diffusion; it preferentially accumulates in cancer cells and colon cancer tissues compared with normal cells and tissues; it mainly localizes to mitochondria, with a small amount also distributed in the cytoplasm and cell membrane. AG2 can be used for colon cancer research[1].

In Vitro

AG2 functions as a polarity-sensitive, pH-insensitive two-photon tracer with a high Φδ value of 88 GM at 780 nm in PBS buffer[1].
AG2 (6-100 μM; 30 min) exhibits negligible cytotoxicity in A549 cells, with 50 μM identified as the optimal concentration for cellular uptake experiments, as higher concentrations induce cell death[1].
AG2 (50 μM; 10 min) localizes predominantly to mitochondria and cytosol/membrane regions in A549 cells, with cytosolic AG2 selectively detectable via a 520-620 nm emission window[1].
AG2 (50 μM; 10 min) is selectively taken up by A549 and HeLa cancer cells over HEK293 and NIH/3T3 non-cancer cells, and its uptake in A549 cells is mediated by a glucose-specific transport system, as shown by competitive inhibition with d-glucose but not l-glucose[1].
AG2 (50 μM; 10 min) uptake by A549 cells is inhibited in a time- and dose-dependent manner by taxol and combretastatin, demonstrating its utility for screening anticancer agents via TPM[1].
AG2 (50 μM; up to 4000 s or 4 h) is preferentially taken up by human colon cancer tissue slices over normal tissue slices, with uptake inhibited by taxol pre-treatment, enabling visualization of glucose metabolism at depths of 75-150 μm for over 3000 s via TPM[1].
