Alizarin red S indicator (C.I. 58005) is a reductively active (quinone-based) anthraquinone dye that forms complexes with metal ions (such as zirconium, calcium) or boric acid to label calcium deposition and perform electrochemical sensing functions. Alizarin Red S undergoes reversible redox reactions (for electrochemical detection) and irreversible chelation (for bone staining). Alizarin Red S is mainly used in bone metabolism research (labeling mineralized tissue), sugar detection (boric acid-sugar competition system) and metal ion sensing (such as zirconium ion detection), and can be used in osteoporosis and metabolic disease research.

Biological Activity
Alizarin Red S (0.1 mM) is used as an electrochemical indicator in cyclic voltammetry. Through interaction with Phenylboronic Acid (HY-W001090) and the competitive effect of fructose, fructose is detected with a linear range of 50 mM[1].
Alizarin Red S (0.05 M; 5 min) is immobilized on a triacetylcellulose membrane after forming an ion pair with hexadecyltrimethylammonium bromide (CTAB) to construct a zirconium ion optical sensor. The detection range of Zr[4+] is 0.39-301 μg/mL, and the detection limit is 0.08 μg/mL[2].

Animal Model:Bone Staining in Wistar Rats Animal Model: Wistar albino rats (4-120 days old, mixed gender)[3]

Dosage:100 mg/kg (4-60 days) or 125 mg/kg (≥60 days) in 0.45% NaCl solution

Administration:Single intraperitoneal injection; sacrificed 2 min-72 h post-injection.

Result:Rapidly absorbed (<2 min) by all bone surfaces, forming fluorescent lines. In 4-60-day-old rats, broader lines indicated active apposition, while thinner lines marked resorption sites.
Resulted complete cortical table staining, suggesting immature mineralization in younger rats (14 days), whereas displayed only surface labeling in older rats (≥60 days).
Irreversibly on appositional surfaces but removed by resorption, confirming its utility for tracking dynamic bone remodeling.

- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)

This protocol yields a clear solution of ≥ 1.25 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (12.5 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
