Bis(2-ethylhexyl) adipate
Based on 1 Customer Validation
Bis(2-ethylhexyl) adipate (NSC 56775) is an aliphatic plasticizer with certain toxicity. Bis(2-ethylhexyl) adipate is a rodent hepatic peroxisome proliferator that dose-dependently increases relative liver weight in rodents, upregulates palmitoyl-CoA oxidase (PCO) activity, and induces hepatocyte hypertrophy and glycogen loss. Long-term exposure to Bis(2-ethylhexyl) adipate induces hepatocellular tumors in B6C3F1 mice. Bis(2-ethylhexyl) adipate induces maternal gestational toxicity and reduced offspring birth weight in a prenatal-postnatal exposure Wistar rat model. Bis(2-ethylhexyl) adipate can be used in metabolism-related research and organic synthesis-related research.

Biological Activity
Description
In Vitro
Bis (2-ethylhexyl) adipate (DEHA) can be metabolized by human liver microsomes to produce adipic acid and MEHA, and initially generate MEHHA/MEOHA[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.
In Vivo
Bis (2-ethylhexyl) adipate (0.5-2.5 g/kg/day; gavage; once daily; 14 consecutive days) dose-dependently increases relative liver weight, PCO activity, peroxisomal volume, and the PCO/catalase ratio in Fischer 344 rats and B6C3F1 mice; male Fischer 344 rats are more sensitive to peroxisome induction than B6C3F1 mice[2].
Bis (2-ethylhexyl) adipate (200-1200 mg/kg/day; p.o.; daily; from GD 7 to postnatal day 17 of the offspring) can cause a dose-related increase in postnatal mortality, prolong gestation, and reduce offspring body weight, with a NOAEL for developmental toxicity of 200 mg/kg, but it does not produce anti-androgenic effects[3].

Dosage:0, 0.5, 1.0, 1.5, and 2.5 g/kg/day
-
Administration:gavage; once daily; 14 consecutive days
-
Result:Dose-dependently increased relative liver weight, PCO activity, peroxisomal volume, and the PCO/catalase ratio in Fischer 344 rats and B6C3F1 mice; male Fischer 344 rats are more sensitive to peroxisome induction than B6C3F1 mice.

Animal Model:Wistar ( time-mated nulliparous young adult females approximately 200 g)[3]
-
Dosage:200 mg/kg/day, 400 mg/kg/day, or 800 mg/kg/day
-
Administration:p.o. (gavage); daily; from GD 7 to pup age day 17
-
Result:Reduced maternal gestational weight gain from GD 7‑21 and elevated perinatal loss in Wistar rats at doses ≥400 mg/kg/day.
Decreased male and female pup birth weight in a dose‑dependent manner, without altering gestation length or male pup sex ratio after chemical treatment.
Increased postnatal death rates and the proportion of affected litters at 400 mg/kg/day and 800 mg/kg/day upon chemical exposure.
Lowered offspring body weight at PND 3 and PND 13 in the 800 mg/kg/day group, but induced no changes in male anogenital distance, male nipple count, timing of vaginal opening or preputial separation.
Elevated relative liver weight at PND 21 under the highest‑dose exposure, while produced no obvious weight shifts for most juvenile reproductive organs and testicular testosterone levels.
Decreased adult terminal body weight and paired adrenal weight at 800 mg/kg/day, and exerted negligible influences on adult reproductive‑organ weight, sperm kinematics, serum prolactin and reproductive‑organ histopathology.
