HKSOX-1 (5/6-mixture) is a fluorescent probe which is used for imaging and detection of endogenous superoxide in live cells and in vivo. HKSOX-1 (5/6-mixture) exhibits excellent selectivity and sensitivity towards superoxide anion radical (Ex/Em = 509/534 nm).

1. Solution preparation
1.1 Preparation of stock solution
Solvent: DMSO
Concentration: 10 mM is recommended.
1.2 Preparation of working solution
Dilute to 10 μM with phosphate buffer (optimized according to the experiment).
Note: The working solution should be prepared and used immediately. Keep it away from light.
2. Superoxide staining
2.1 The reaction of HKSOX-1 (10 μM) with O2?? (10 equiv, 100 μM) generated by enzymatic reaction of xanthine (300 μM) and xanthine oxidase (0.01 U/mL) in potassium phosphate buffer (0.1 M, pH 7.4) at 25°C.
2.2 The fluorescence intensity is monitored at the Ex/Em = 509/534 nm.
