DOPE-PEG3400-pPB (DOPE-PEG3400-CSRNLIDC) is a functionalized PEGylated lipid conjugate composed of three modular components: dioleoylphosphatidylethanolamine (DOPE) as the lipid anchoring group, a polyethylene glycol (PEG) spacer arm with a molecular weight of 3400 Da, and platelet-derived growth factor receptor β-binding peptide (pPB peptide (HY-P11549)) as the terminal targeting/functional ligand. DOPE-PEG3400-pPB enables targeted delivery to fibrotic tissues and tumor stroma.

Biologische Aktivität
DOPE-PEG3400-pPB is specifically designed for active targeting nanomedicine applications. It can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against PDGFRβ on the surface of activated hepatic stellate cells, pericytes, and tumor-associated fibroblasts.
Composition and role of each component:
1. PEG3400 (polyethylene glycol, MW 3400): Hydrophilic polymer spacer arm that forms a steric corona around nanoparticles, reducing opsonization and reticuloendothelial system (RES) clearance, prolonging circulation half-life, and providing spatial separation between the lipid surface and targeting ligand.
2. pPB (platelet-derived growth factor receptor β-binding peptide (pPB)): Targeting ligand-a peptide that specifically recognizes and binds to platelet-derived growth factor receptor beta (PDGFRβ), which is highly expressed on activated hepatic stellate cells (HSCs), pericytes, and tumor-associated fibroblasts. Enables targeted delivery to fibrotic tissues and tumor stroma.
