DMG-PEG5000-cRGD

DMG-PEG5000-cRGD is specifically designed for active targeting nanomedicine applications. It can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against αvβ3/αvβ5 integrins on tumor vasculature and cancer cell surfaces.
Composition and role of each component:
1. DMG (dimyristoylglycerol): Lipid anchoring group-a diacylglycerol with two C14 chains, providing moderate membrane anchorage; its short-chain, high-fluidity characteristics enable faster dissociation of PEG-lipids from nanoparticle surfaces, facilitating endosomal escape and cellular uptake.
2. PEG5000 (polyethylene glycol, MW 5000): Hydrophilic polymer spacer arm that forms a steric corona around nanoparticles, reducing opsonization and reticuloendothelial system (RES) clearance, prolonging circulation half-life, and providing spatial separation between the lipid surface and targeting ligand.
3. cRGD (cyclic RGD peptide (cyclo(Arg-Gly-Asp))): Targeting ligand-a cyclic peptide containing the arginine-glycine-aspartic acid (RGD) motif that selectively binds to αvβ3 and αvβ5 integrins, which are highly expressed on tumor neovasculature, activated endothelial cells, and certain cancer cells.
