Emugrobart (GYM-329, RG6237, RG-70240) is a humanized IgG1κ antibody targeting myostatin (Myostatin; GDF8). Emugrobart binds to pro-myostatin and latent myostatin, blocking their cleavage into mature myostatin; it also has a clearance function, which transports bound myostatin for degradation and allows for cyclic reuse. Emugrobart enhances muscle strength in mouse models of muscle atrophy and increases muscle mass in cynomolgus monkeys. Emugrobart can be used in research on spinal muscular atrophy and facioscapulohumeral muscular dystrophy.

Emugrobart incorporates a feature termed the "clearance function": after the formation of antibody-antigen complexes, such complexes are internalized by specific endothelial cells and immune cells. Inside the cells, the complexes dissociate; the free myostatin is directed to degradation, while the free antibodies are recycled, redirected to the cell surface and released into the extracellular space to exert their effects again[1].

Flow cytometric analysis of 1X106 HeLa cells with Emugrobart (HY-P990993, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. AF 488-conjugated AffiniPure Goat Anti-Human IgG H&L (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa (HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
