Fmoc-Tic-OH, an Fmoc-protected derivative of Tic, is a solid-phase peptide synthesis monomer. Fmoc-Tic-OH protects the amino group of Tic via a base-labile Fmoc group, allowing conformationally constrained Tic residues to be stepwise coupled to peptide chains, and the Fmoc protecting group can be removed under piperidine conditions. Fmoc-Tic-OH can be used for peptide synthesis and opioid receptor ligand research.

Biological Activity
Fmoc-Tic-OH is used in standard Fmoc solid-phase peptide synthesis. In the synthesis of TIPP biotinylated derivatives, Fmoc-Tic-OH is coupled to the growing peptide chain using DIC/HOBt, followed by subsequent coupling of Boc-Tyr (OtBu)-OH, ultimately yielding the biotinylated Tyr-Tic-Phe-Phe-Asp peptide containing a Tic residue[1].
Fmoc-Tic-OH is used in the Fmoc/tBu solid-phase synthesis of H-Dmt-Tic-Xaa series peptides. After the coupling of the C-terminal amino acid, Fmoc-Tic-OH is added and coupled with HATU (4 eq) in DMF for 1 h, followed by subsequent coupling with Boc-Dmt-OH to construct the Dmt-Tic pharmacophore[2].
