Induction of Arthritis[2]
Background
G6PI is capable of producing antibodies in the body that induce arthritis by inducing immunoactive cells to produce cytokines (such as TNF-α, IL-1β).
Specific Modeling Methods
Mice: DAB/1 • 6-10 week old
Administration: 50 μg in complete Freund's adjuvant • s.c. at the base of the tail • single dose
Note
(1) DBA/1 mice were bred and maintained under specific-pathogen free conditions in our animal facility.
(2) Mice were sacrificed on day 12 after immunization and single-cell suspensions from draining lymph nodes were prepared.
Modeling Indicators
Morphology: Showed a strong synovialitis and inflammation of the adjacent skeletal muscle , which was dominated by neutrophilic granulocytes as a sign of acute inflammation and exhibited activation of synovial fibroblasts to a lesser extent. The synovial tissue was enlarged due to the dense inflammatory infiltrate; however, bone destruction was onlyfocally observed.
Molecular changes: Increased the levels of IL-17, TNFα, RANKL, and IFNγ.
