SR-4559 is a α/β hydrolase domain-containing protein 5 (ABHD5) ligand. SR-4559 activates adipose triglyceride lipase (ATGL) and stimulates lipolysis by inhibiting ABHD5-PLIN interactions in adipocytes and muscle cells. SR-4559 can be used for metabolic diseases like Chanarin Dorfman syndrome research.

SR-4559 (10-9-10-4 M; 4 h) potently inhibits the interaction between ABHD5 and PLIN1 (IC50 = 510 nM) in a cell-free luciferase complementation assay[1].
SR-4559 (10-9-10-4 M; 4 h) potently and equally disrupts the interaction between ABHD5 and PLIN5 in a cell-free luciferase complementation assay[1].
SR-4559 (1 μM; 0-1 min) rapidly dissociates the PLIN1(358-415)-ABHD5 complex on lipid droplets in Cos7 cells within 1 minute[1].
SR-4559 stimulates lipolysis in brown adipocytes with an EC50 of 4 μM[1].
SR-4559 (10 μM) requires ABHD5 to stimulate lipolysis in brown adipocytes, as activity is abolished by ABHD5 knockdown and restored by ABHD5 rescue[1].
