Hercynylcysteine sulfoxide

Hercynylcysteine sulfoxide is a synthetic precursor of Ergothioneine (HY-N1914), and it spontaneously converts to Ergothioneine in the presence of pyridoxal phosphate (PLP). Hercynylcysteine sulfoxide is produced via the fusion enzyme complex NcEgt1‑1/CtEgtB-catalyzed conversion of L-histidine and L-cysteine, and serves as an intermediate in the ergothioneine biosynthetic pathway of Schizosaccharomyces pombe and engineered Escherichia coli BL21 (DE3).

Hercynylcysteine sulfoxide is undetectable in wild-type Schizosaccharomyces pombe cells under vegetative growth conditions; it accumulates in egt2Δ deletion mutant Schizosaccharomyces pombe cells cultured under vegetative growth, nitrogen starvation, or glucose starvation conditions for 24 hours^[1].
Hercynylcysteine sulfoxide accumulates in Schizosaccharomyces pombe egt1⁺ overexpression strains in a promoter strength-dependent manner under thiamine-free culture conditions; it accumulates in large quantities in Schizosaccharomyces pombe P3nmt1-egt1⁺ Δegt2 double mutant cells cultured in selenium-supplemented medium^[1].
Hercynylcysteine sulfoxide is a transient biosynthetic intermediate produced by engineered Escherichia coli BL21 (DE3) via a two-step consecutive enzymatic reaction involving NcEgt1-1 and CtEgtB^[2].
