h-FTAA
Based on 1 Customer Validation
h-FTAA is a luminescent conjugated oligothiophene amyloid-binding probe. h-FTAA binds pre-fibrillar amyloid-β (Amyloid-β) and AβArc aggregates, promotes fibril/protofibril formation, accelerates amyloid-β fibrillation, and reduces amyloid-β and AβArc-induced cytotoxicity and neurotoxicity-associated morphological changes in neuronal cells. h-FTAA recognizes the periphery of immature plaques, postmortem human brain plaque pathology, and all familial brain tissue Aβ deposits, labeling both plaque populations in a pan-amyloid binding manner. h-FTAA stains the corona of cored plaques and diffuse plaques, enabling multiplexed hyperspectral imaging. h-FTAA can be used for research on Alzheimer's disease.

Biological Activity
Description
In Vitro
h-FTAA (3 μM; 25 min) labels the diffuse immature peripheral regions of Aβ plaques in ex vivo brain sections from tgSwe and tgArcSwe mice[2].
h-FTAA (1.5 μM; 30 min) fluorescently labels Aβ plaques in postmortem PSEN1 fAD temporal cortex and validates MALDI-MSI plaque ROIs[3].
h-FTAA (200 nM; 30 min at room temperature) fully co-localizes with 4G8 and labels all Aβ deposits in PSEN1 A431E frontal cortex sections, including both the HS-276-positive and LL-1-positive plaque populations, with no laminar differences[4].
h-FTAA (3 μM; up to 3 h) attenuates Aβ1-42-induced toxicity in SH-SY5Y neuroblastoma cells upon co-aggregation[5].
h-FTAA (10 μM; aggregation 0-4 h; exposure 72 h) completely eliminates AβArc-mediated cytotoxicity in SH-SY5Y neuroblastoma cells, whereas 1 and 0.1 μM are inactive[5].
h-FTAA (10 μM; 0 h) prevents Aβ1-42- and AβArc-induced neurotoxic morphology in SH-SY5Y neuroblastoma cells[5].
h-FTAA (3-10 μM; Aβ1-42 3 h; AβArc 0 h) alters the morphology of Aβ1-42 and AβArc aggregates, among which AβArc forms protofibrils at 0 h[5].
h-FTAA (0.3 μM; 20 h) accelerates Aβ1-42 aggregation with a lag time of 1.39 h and a t0.5 of 2.47 h, whereas AβArc aggregation remains non-sigmoidal within 20 h[5].
h-FTAA (0.3 μM; 0-25 h) binds to Aβ1-42, shifting the excitation wavelength from 450 nm to 480 nm, and rapidly binds AβArc aggregates, reaching a binding plateau at 10 h[5].

Cell Line:SH-SY5Y human neuroblastoma cells
-
Concentration:3 μM (h-FTAA); 10 μM (Aβ1-42 aggregation); 3 μM (Aβ1-42 cell exposure)
-
Incubation Time:1, 2, 3, 5 h (aggregation at 37°C); 72 h (cell exposure)
-
Result:Co-aggregation increased SH-SY5Y viability relative to Aβ1-42 alone at the early aggregation times; the protective effect was reported for cell exposures after ≤ 3 h of Aβ1-42 aggregation.

Cell Line:SH-SY5Y human neuroblastoma cells
-
Concentration:0.1, 1, 10 μM (h-FTAA); 10 μM (AβArc aggregation); 3 μM (AβArc cell exposure)
-
Incubation Time:1, 2, 4 h (aggregation at 37°C); 72 h (cell exposure)
-
Result:At 10 μM, a protective effect was observed at all investigated times (1, and 2 h).
At an equimolar AβArc concentration, the cytotoxic effect was completely abolished.
Lower concentrations (0.1 and 1 μM) did not protect after any exposure period.
