Isostrictiniin
Isostrictiniin (ITN) is a polyphenolic compound isolated from Nymphaea candida. It upregulates the expression of Nrf2 and HO-1, while downregulating the expression of Keap1. Isostrictiniin reduces the phosphorylation levels of JNK, ERK1/2, p38, IκBα, and NF-κB p65. Isostrictiniin decreases the levels of pro-inflammatory mediators (IL-1β, IL-6, TNF-α, PGE2). Isostrictiniin alleviates LPS-induced acute lung injury and exerts protective effects against acute alcoholic liver injury. Additionally, isostrictiniin exhibits anti-inflammatory, antioxidant, and anti-fibrotic activities.

Isostrictiniin (50-200 μM; 12 h) impairs proliferation of A549 cells, reducing cell survival rate below 80%[2].
Isostrictiniin (25 μM; 12 h) reduces pro-inflammatory mediator levels (IL-1β, IL-6, TNF-α, PGE2) in LPS-induced A549 cells[2].
Isostrictiniin (10-25 μM; 12 h) upregulates Nrf2 and HO-1, and reduces Keap1 in LPS-induced A549 cells, regulating the Keap1-Nrf2/HO-1 pathway[2].

Reduced lung tissue myeloperoxidase (MPO) activity (p < 0.05), lung tissue tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) levels (p < 0.01) at 50, 100 mg/kg. Suppressed blood neutrophil counts, elevated lymphocyte counts (p < 0.01), reduced lung tissue malondialdehyde (MDA) levels, and increased superoxide dismutase (SOD) and glutathione (GSH) levels (p < 0.05) at 100 mg/kg. Improved lung histopathological damage and reduced pathological scores (p < 0.05) at 25, 50, 100 mg/kg.
