Guidelines (The following is our recommended experimental protocol. This protocol serves only as a reference guide, and specific operations should be adjusted according to your actual needs.)
Observation of New Linear Structures in Rabbit Lymphatic Vessels by Janus Green B Staining[1]
(1) Experimental Animal Preparation: Anesthetize 12-week-old New Zealand rabbits systemically, make an incision on the left or middle part of the rabbit's abdomen, and expose the large lymphatic vessels adjacent to the inferior vena cava.
(2) Staining: Use a No. 30.5 fine needle to slowly inject approximately 0.3 mL of pre-warmed Janus Green B staining solution into a lymph node or lymphatic vessel near the inferior vena cava as slowly as possible. Gently withdraw the needle and observe the blue color presented by the flowing Janus Green B staining solution in the lymphatic vessels.
(3) Washing and Incubation: Inject phosphate-buffered saline (pH 7.4) pre-warmed to 40°C to remove the retrograde Janus Green B staining solution, and maintain tissue temperature and viability.
(4) Observation and Localization: Carefully observe the internal condition of the lymphatic vessels using a stereomicroscope, and determine the position of well-stained regions. Adjust the focus of observation according to staining effects and individual differences.
(5) Sample Fixation: Immediately immerse the harvested samples in 10% neutral formalin solution for fixation.
(6) Additional Staining (Optional): Select some fixed samples and perform secondary staining with 0.1% (w/v) acridine orange PBS solution.
(7) Imaging: Perform imaging analysis of the stained samples using a confocal laser scanning microscope.
