Allantoxanamide (250 mg/kg; i.p.) induces hyperuricemia in rats by inhibiting uricase, increasing serum uric acid levels to 14 mg/dL at 6 h after administration, with elevated levels maintained at 24 h^[1].
Allantoxanamide (150 mg/kg; i.p.) induces acute hyperuricemia in Sprague-Dawley rats and reduces renal 1-α hydroxylase protein and 1,25(OH)₂D levels^[2].
Allantoxanamide (150 mg/kg; i.p.) produces sustained hyperuricemia in rats, accompanied by reduced uric acid excretion, renal dysfunction, proteinuria, and urinary occult blood^[3].

Allantoxanamide inhibits uricase, blocking the degradation of uric acid to allantoin and inducing elevated serum uric acid levels in rats^[1]^[3].

Specific Modeling Methods

Rats: Male•Slc-Wistar rats•(9-10 weeks old)
Administration: Allantoxanamide (150 mg/kg; i.p.; once) suspended in 1% gum arabic solution (2 mL/kg body weight) to induce hyperuricemia

(1) Allantoxanamide was administered as a suspension in 1% gum arabic solution at a volume of 2 mL/kg body weight.
(2) Allantoxanamide-induced hyperuricemia is associated with reduced renal uric acid excretion and renal dysfunction due to nephrotoxicity in addition to uricase inhibition.
