Description

Lipid C16-C is an ionizable cationic lipid that can be used to prepare lipid nanoparticles (LNPs) for mRNA delivery. Lipid C16-C mediates ApoE-independent macropinocytosis-based mRNA delivery to macrophages, with endosomal acidification enabling endosomal escape and release of mRNA cargo into the cytosol[1].

In Vitro

Lipid C16-C (250-500 ng mRNA per 50k cells; 24 h) enables potent mRNA delivery to PMA-differentiated human THP-1 macrophages, with the optimized B15 formulation showing 13-fold enhanced delivery relative to the initial formulation[1].
The B15 formulation with Lipid C16-C (up to 1500 ng mRNA per 50k cells; 24 h) enables robust mRNA delivery to both GMCSF- and MCSF-derived primary human macrophages, with differential transfection profiles between the two models[1].
