Mag-indo-1/AM is a cell-permeable fluorescent indicator for Mg2+ and also for Ca2+ (Ex=340-390 nm, Em=410-490 nm). Mag-indo-1/AM can be used to determine intracellular Mg2+ and Ca2+ concentrations.

Biological Activity
Guide (The following is the experimental plan we recommend. This plan serves only as a reference guide. The specific operations should be adjusted according to your actual needs.)
Measurement of intracellular Ca2+ content[1]:
1. Take 3 mL of cell suspension, add 10 μM Mag-indo-1/AM, and incubate at 30°C for 15 minutes.
2. Centrifuge at 500 rpm for 1 minute, resuspend the cells in 5 mL of separation buffer (containing 1 mM CaCl2?).
3. Add 1 mM Probenecid (HY-B0545) to the cell suspension to prevent the dye from leaking out of the cytoplasm; place the cells loaded with Mag-indo-1/AM at room temperature for standby.
4. Place the cells loaded with Mag-indo-1/AM at room temperature for 2 hours, then gently shake them at 37°C for 1.5 hours; centrifuge to collect the cell precipitate, resuspend and store at room temperature.
5. Take a small amount of the loaded dye cells and place them in an experimental chamber. Place the chamber on the stage of an inverted microscope.
6. Detect the fluorescence signal of Mag-indo-1/AM (excitation wavelength Ex = 340-390 nm, emission wavelengths Em = 410 nm and 490 nm, which are the emission peaks indicating the bound and free Ca2+ states of the indicator). Collect the full-cell fluorescence signal in real time using a photon counting system.
