Met-Gly-Pro-AMC is a fluorescent peptide substrate active against *Caenorhabditis elegans* DPF-3, hDPP4, and methionyl aminopeptidase 2 (MetAP2). Met-Gly-Pro-AMC undergoes cleavage via tripeptidyl peptidase activity to release a fluorescent product. Met-Gly-Pro-AMC is used to detect the activities of tripeptidyl peptidases and MetAP2 peptidases in in vitro enzyme activity assays.

The fluorogenic tripeptide substrate Met-Gly-Pro-AMC (0.5-600 μM; 30 min) is robustly processed by recombinant human DPP4 (Km = 461.5 μM), moderately processed by recombinant C. elegans DPF-3 (Km = 245.6 μM), and not processed by recombinant human DPP9 or catalytically impaired DPF-3S784A[1].
