Myokinasib-II
Myokinasib-II is a MLCK1 inhibitor with an IC50 of 4.4 μM. Myokinasib-II induces cell cortex softening, impairs cell migration capacity, induces multinucleation, reduces cell stiffness, alters membrane tether dynamics, decreases actomyosin tension and changes nuclear morphology. Myokinasib-II is a MLCK1 chemical biology probe with a unique structure that does not depolymerize tubulin and maintains cell viability even under high-concentration treatment in fibroblasts.

Biological Activity
|
MLCK1 4.4 μM (IC50) |
Myokinasib-II (10 μM) reduces the fluorescence intensity of pMLC without disrupting actin stress fibers in NIH3T3 mouse embryonic fibroblasts[1].
Myokinasib-II (10 μM; 6-24 h) reduces cortical stiffness and nucleomyosin tension in NIH3T3 cells, inhibits cell migration, and induces multinucleation[1].
