Naphthyridine carbamate dimer (NCD) is a molecular glue. Naphthyridine carbamate dimer induces designed structural changes in target functional DNA, thereby regulating its function. As a molecular glue, naphthyridine carbamate dimer stabilizes non-spontaneously formed GG homologous mismatched double-stranded DNA. Naphthyridine carbamate dimer reduces the formation of toxic RNA foci and regulates CGG repeat instability associated with fragile X syndrome and spinocerebellar ataxia type 31. Naphthyridine carbamate dimer can be used in research related to fragile X syndrome and spinocerebellar ataxia type 31.

In Vitro
Naphthyridine carbamate dimer (NCD) (20 μM) increases the melting temperature of GG1 double-stranded DNA from 26.8°C to 48.6°C, enhancing the thermodynamic stability of the duplex[1].
Naphthyridine carbamate dimer binds to GG1 hairpin DNA and d(CGG)10 DNA with a 2:1 NCD:DNA stoichiometry, as confirmed by detection of complex ions with two NCD molecules per DNA molecule[1].
Naphthyridine carbamate dimer binds to GG1 double-stranded DNA with an apparent KD of 67 nM and a 2:1 NCD:DNA stoichiometry, with binding driven by enthalpic contributions[1].
Naphthyridine carbamate dimer (1:2 molar ratio of RNA to NCD) binds to the UGGAA-repeat-containing ROG1 and ROG2 RNA duplexes, inducing a Z-RNA-like structural rearrangement of the UGGAA motif and acting as a molecular glue to stabilize crystal lattice formation in the ROG1-NCD complex[3].
Naphthyridine carbamate dimer binding induces measurable conformational changes in the ROG2 RNA duplex, as detected by circular dichroism spectroscopy[3].

- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 1.25 mg/mL (2.48 mM); Clear solution
This protocol yields a clear solution of ≥ 1.25 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (12.5 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
