M918
M918 is a cell-penetrating peptide derived from the C-terminal region of the tumor suppressor p14ARF. Containing multiple arginine residues, M918 can bind to protein cargo and antisense PNA. By facilitating the delivery of sequence-specific antisense PNA, M918 inhibits and corrects splicing-thereby silencing luciferase expression-while simultaneously inducing Th1-type immune responses, including the production of IFN-γ, IgG2a, IgG2b, and Granzyme B. The intracellular internalization of M918 is independent of glycosaminoglycans; it exhibits low cytotoxicity and can be utilized in research concerning HIV-1 infection and related delivery strategies.

Biological Activity
Description
In Vitro
M918 (1 µmol/l; 30 min) partially co-localizes with both transferrin and dextran in MCF-7, CHO, and Hifko cells, but not with transferrin in HeLa cells[5].
M918 internalization is independent of glycosaminoglycans on the cell surface in CHO cells[5].
M918-PNA (5 µmol/l; 1 h) induces splice correction dose-dependently in HeLa pLuc 705 cells, with a 15-fold increase at 5 µmol/l[5].
M918 forms stable, non-covalent nanoparticles with Nef and Hsp20-Nef proteins; these nanoparticles enter cells via clathrin-mediated endocytosis and macropinocytosis, and exhibit robust endosomal escape and protein transduction capabilities[3].
M918 (5-25 µmol/l; 72 h) does not significantly affect cellular proliferation after 72 hours in any tested cell line[5].
M918 (1 µmol/l; 30 min) displays mainly vesicular localization inside MCF-7, Hifko, and HeLa cells[5].
M918 (1-5 µmol/l) is the most efficient vector for delivering streptavidin and avidin into cells when utilizing biotinylated peptides at 1 µmol/l, and it also promotes uptake of streptavidin at 5 µmol/l as a non-covalent complex[5].
