p-Nitrobenzyl mesylate (4-Nitrobenzyl mesylate) is an alkylating reagent. p-Nitrobenzyl mesylate alkylates thiophosphorylated serine, threonine, tyrosine residues and cysteine thiols, without modifying hydroxyl, amino or carboxylic acid functional groups. p-Nitrobenzyl mesylate generates thiophosphate epitopes on thiophosphorylated substrate proteins for recognition by thiophosphate-specific antibodies. p-Nitrobenzyl mesylate facilitates the identification and purification of kinase substrates. p-Nitrobenzyl mesylate is applicable to studies related to cerebral ischemia-reperfusion injury.

Biological Activity
p-Nitrobenzyl mesylate (2.5 mM; 1 h) achieves complete monoalkylation of a model thiophosphopeptide[1].
p-Nitrobenzyl mesylate alkylates cysteine residues and thiophosphorylated serine residues in c-Jun-GST, but does not react with alcohol, amine, or carboxylic acid functional groups in the protein[1].
p-Nitrobenzyl mesylate (2.5 mM; 2 h) effectively alkylates thiophosphorylated Arabidopsis substrates VQ4 and MKS1, allowing specific detection via anti-TPE antibody to confirm that mutation of MPK gatekeeper residues to alanine does not alter substrate specificity of Arabidopsis MPK3, MPK4, and MPK6[3].
p-Nitrobenzyl mesylate (2.5 mM; 2 h) effectively alkylates thiophosphorylated Arabidopsis substrates AT1G78150, VQ4, and MKS1, allowing specific detection via anti-TPE antibody to confirm differential substrate preferences of Arabidopsis AS-MPK3, AS-MPK4, and AS-MPK6[3].
