BDCA-2

BDCA-2 (CLEC4C) is a defining surface marker of human plasmacytoid dendritic cells and a therapeutic target in interferon-driven autoimmune disease. This type II transmembrane C-type lectin lacks a signaling tail of its own and couples to the Fc receptor gamma-chain; importantly, its engagement transmits an inhibitory signal that blocks TLR-induced type I interferon and chemokine production. It also functions as an endocytic receptor, internalizing bound antigens for presentation. Because plasmacytoid dendritic cells are major interferon-alpha producers in systemic lupus erythematosus, BDCA-2-directed antibodies are being developed to deplete or silence these cells. Recombinant BDCA-2 supports pDC biology, lupus research, and biologic-drug screening.
