Biological Activity

Description

Complement Factor B/CFB Protein, Human (HEK293, His) is the recombinant human-derived Complement Factor B/CFB protein, expressed by HEK293, with C-6*His labeled tag.

Background

Complement factor B (CFB) is part of the replacement pathway of the complement system and can be cut by factor D into two fragments: Ba and Bb. Bb is a serine protease, which then binds to complement factor 3b to produce C3 or C5 invertase, and is also associated with the proliferation and differentiation of pre-activated b lymphocytes, rapid diffusion of peripheral mononuclear cells, stimulation of lymphoblastogenesis, and lysis of red blood cells. Ba inhibits the proliferation of preactivated b lymphocytes. Activation of the complement replacement pathway (AP) is thought to play an important role in immunoglobulin A nephropathy (IgAN). CFB can inhibit the activity of AP. The expression of CFB was positively correlated with T cell CD8, macrophage M1, plasma cells, T cell CD4 memory activation, T cell follicular assist, and T cell regulation (Tregs). It was negatively correlated with eosinophils, macrophages M0, macrophages M2, mast cell rest and T cell CD4 memory rest in TME. CFB regulates cellular senescence and is associated with poor prognosis in pancreatic cancer[1][2][3][4][5][6].

Verified Bioactivity

1.Immobilized Iptacopan (HY-127105) at 20 μg/mL (100 μL/well) can bind Biotinylated Human CFB, the ED50 for this effect is 17.83 nM.
2.Immobilized CFB,Human (HY-P7813) at 2 μg/mL (100 μL/well) can bind Biotinylated Iptacopan, The ED50 for this effect is 11.41 nM.
3.Human CFB Immobilized on CM5 Chip can bind Iptacopan (HY-127105) with an affinity constant of 7.59 nM as determined in SPR assay.
