Background
PRK2 is a PKC-related serine/threonine-protein kinase and Rho/Rac effector protein involved in specific cellular signal transduction responses. It regulates cell cycle progression, actin cytoskeleton assembly, cell migration, adhesion, tumor invasion, and transcriptional activation. PRK2 phosphorylates CTTN in hyaluronan-induced astrocytes, reducing CTTN's F-actin binding capacity, and HDAC5 to impair its nuclear import. As a direct RhoA target, it mediates primordial-to-apical junction maturation in bronchial epithelial cells. It facilitates G2/M phase progression and cytokinesis abscission via ECT2, stimulates FYN kinase activity for keratinocyte adhesion, and modulates bladder epithelial cell migration direction/speed during tumor invasion. PRK2 inhibits Akt pro-survival signaling, mediates Rho-induced transcription via c-fos SRF, and negatively regulates ciliogenesis. by similar: During microbial infection, it phosphorylates HCV NS5B to enhance HCV RNA replication.
