Background
SIRT2 is an NAD-dependent protein deacetylase that targets histones (e.g., H4K16ac, H3K56ac), α-tubulin (e.g., Lys-40), and key transcription factors (e.g., FOXO1, FOXO3, HIF1A, RELA, p53/TP53). It regulates diverse processes including cell cycle control (e.g., G2/M transition, APC/C activation), genomic stability, microtubule dynamics, metabolic pathways (e.g., PPP, gluconeogenesis), autophagy, and epigenetic modifications (e.g., H4K20me1 deposition). During bacterial infection (e.g., Listeria monocytogenes) or oxidative stress, SIRT2 modulates transcriptional activity and stress responses via deacetylation. It also exhibits demyristoylation (e.g., ARF6) and depalmitoylation (e.g., KRAS) activities, influencing membrane localization. by similar: SIRT2 promotes FOXO3 ubiquitination and degradation through deacetylation.
