JTE-607, a highly selective inflammatory cytokine synthesis inhibitor, protects from endotoxin shock in mice. JTE-607 inhibits inflammatory cytokine production, including TNF-α, IL-1β, IL-6, IL-8 and IL-10, from LPS-stimulated human PBMCs, with IC50s of 11, 5.9, 8.8, 7.3 and 9.1 nM, respectively . AN15368 is an orally active trypanocidal agent and a prodrug of AN14667. AN15368 selectively targets CPSF3-mediated mRNA maturation in intracellular amastigotes of Trypanosoma cruzi, reducing the mRNA levels of the parasite. As a prodrug, AN15368 can be cleaved into its active carboxylate form by CBP of Trypanosoma cruzi. AN15368 completely clears long-term naturally acquired Trypanosoma cruzi infections in non-human primates. AN15368 can be used in the research of Chagas disease .

AN11736 is a CPSF3 inhibitor. AN11736 exerts a potent anti-trypsin effect by specifically inhibiting the activity of the trypanosome CPSF3 endonuclease, disrupting the mRNA maturation process. AN11736 exhibits IC50s against T. congolense and T. vivax of 0.14 and 1.3 nM, respectively. AN11736 exhibits strong anti-trypsin activity in mouse and cattle models .
