Cat. No.: HY-P99529 Purity: 99.00%
Technical Support

Valziflocept (BAX1810; TAK-752) is a recombinant soluble human FcγRIIb receptor that targets Fc and FcγR. FcγRs bind to and neutralize pathogenic IgG, thereby acting as a "decoy" or "scavenger" receptor that reduces the severity of autoimmune diseases. Valziflocept is used in research on autoimmune diseases such as systemic lupus erythematosus (SLE).

Description

Valziflocept (BAX1810; TAK-752) is a recombinant soluble human FcγRIIb receptor that targets Fc and FcγR. FcγRs bind to and neutralize pathogenic IgG, thereby acting as a "decoy" or "scavenger" receptor that reduces the severity of autoimmune diseases. Valziflocept is used in research on autoimmune diseases such as systemic lupus erythematosus (SLE)[1][2][3].

In Vitro

Valziflocept (20 min incubation with opsonised MRBCs; 60 min co-culture with macrophages) significantly inhibits phagocytosis of IgG-opsonised murine red blood cells by thioglycolate-elicited peritoneal macrophages from female NZB/NZW F1 mice[2].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

In Vivo

Valziflocept (200 μg per dose; i.v.; 5 occasions with 3-day intervals) significantly ameliorates collagen-induced arthritis in male DBA/1 mice, reducing mean arthritic scores, pathogenic antibody levels, and inflammatory cytokine mRNA expression[1].
Valziflocept (50-150 μg/week; s.c.; weekly; 4 weeks (prophylactic)) delays onset of lupus pathology and improves survival in NZB/NZW F1 mice, while valziflocept (100 μg/week; s.c.; weekly; until death or study end (therapeutic)) significantly prolongs survival despite established kidney damage[2].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model: DBA/1 (male, age-matched, collagen-induced arthritis model)[1]
Dosage: 200 μg per dose
Administration: i.v.; 5 occasions with 3-day intervals
Result: Reduced mean arthritic score to 6.1 at 12 days after first treatment and 14.2.
Reduced serum IgG anti-collagen type II antibody levels significantly 2 weeks after treatment start.
Reduced TNF-α and IL-6 mRNA levels significantly in lymph nodes.
Reduced TGF-β1, TGF-β2, TGF-β3, and MIF mRNA levels in synovial tissue.
Animal Model: NZB/NZW F1 (female, 5-8 weeks old at study start)[2]
Dosage: 150 μg/week (prophylactic, single weekly dose); 50 μg/week ×3 (prophylactic, three weekly doses); 100 μg/week (therapeutic, single weekly dose until death or study end)
Administration: s.c.; weekly; 4 weeks (prophylactic); s.c.; weekly; until death or study end (therapeutic)
Result: Significantly delayed onset of proteinuria with 3×50 μg/week prophylactic treatment.
Prolonged survival by 11.5 weeks with 3×50 μg/week prophylactic treatment.
Delayed weight loss and delayed development of anaemia with 3×50 μg/week prophylactic treatment.
Reduced histopathological signs of glomerulonephritis, and suppressed mesangial matrix expansion, Bowman space proliferation, and crescent formation in kidneys with 3×50 μg/week prophylactic treatment.
Showed a non-significant trend towards prolonged survival with 1×150 μg/week prophylactic treatment.
Significantly prolonged survival with 1×100 μg/week therapeutic treatment.
Reduced histopathological signs of glomerulonephritis, and suppressed mesangial matrix expansion, Bowman space proliferation, and crescent formation in kidneys with 1×100 μg/week therapeutic treatment.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
