Preparation of Resorufin sodium salt staining solution
1.1 Preparation of stock solution
Prepare a 1000 μM Resorufin sodium salt stock solution in DMSO.
Note: (1) It is recommended to aliquot the Resorufin sodium salt stock solution and store at -20°C protected from light.
1.2 Preparation of working solution
Prepare a 0.4 μM Resorufin sodium salt working solution in PBS (pH 7.4).
Resorufin sodium salt staining
1. Pre-treat HepG2 cells with CD for 30 min, then remove the culture medium. After that, wash the cells three times with 200 μL PBS, and add 150 μL fresh DMEM and 15 μL of 0.4 μM Resorufin sodium salt solution (diluted in DMEM) to each well. Incubate at 37 °C in the dark for 30 min, then measure Resorufin content using a plate reader.
2. Pre-treat HepG2 cells with CD for 120 min, then remove the culture medium. After that, wash the cells three times with 200 μL PBS, and add 150 μL fresh DMEM and 15 μL of 0.4 μM Resorufin sodium salt solution (diluted in DMEM) to each well. Incubate at 37 °C in the dark for 120 min, then measure Resorufin content using a plate reader.
3. Measure fluorescence/absorbance and perform routine quantitative analysis.
