Antifungal agent 180
Antifungal agent 180 is a pyrazole-oxadiazole-linked 1,2,4-triazole derivative and a broad-spectrum antifungal agent. Antifungal agent 180 disrupts fungal cellular homeostasis through multiple pathways, damaging the integrity of fungal hyphal cell membranes, increasing membrane permeability, and causing leakage of intracellular nucleic acids and proteins; it induces hyphal shrinkage, collapse, and organelle disintegration; and it causes widespread dysregulation of sterol/lipid metabolism, carbohydrate utilization, mitochondrial respiration, and stress response pathways at both the transcriptomic and proteomic levels. Antifungal agent 180 exhibits protective and curative efficacy against rice blast in vivo. Antifungal agent 180 can be used in research on rice blast and gray mold.

Description
In Vitro
Antifungal agent 180 (compound L22) exhibits potent and broad-spectrum in vitro antifungal activity against nine plant pathogenic fungi, with the strongest inhibitory effect against Botrytis cinerea (92.7% at 50 μg/mL); and it shows highly effective inhibition against Verticillium dahliae (EC50 = 5.72 μg/mL) and Magnaporthe grisea (EC50 = 5.96 μg/mL)[1].
Antifungal agent 180 increases the cell membrane permeability of Magnaporthe grisea and induces cytoplasmic leakage of nucleic acids and proteins[1].
Antifungal agent 180 (50-100 μg/mL) causes catastrophic structural damage to Magnaporthe grisea hyphae, including surface collapse, cytoplasmic disintegration, and impaired membrane integrity[1].
Antifungal agent 180 (50‑100 μg/mL) results in extensive transcriptional changes and proteomic alterations in Magnaporthe grisea, including downregulation of ergosterol biosynthesis and upregulation of stress response genes; furthermore, at the regulatory level, it disrupts sterol homeostasis, induces oxidative imbalance accompanied by lipid peroxidation, and leads to compensatory metabolic reprogramming[1].

In Vivo
Antifungal agent 180 (1 μg/mL; water immersion; 24 h) is not lethal to zebrafish after 24 h of exposure[1].
