Biological Activity
Antitubercular agent-60 (compound 1) inhibits growth of Mycobacterium tuberculosis H37Rv with an MIC of 3.2 μM in 7H9/DPPC/CAS/Tx media and 3.6 μM in 7H9/GLU/CAS/Tx media[1].
Antitubercular agent-60 disrupts cell wall biosynthesis in Mycobacterium tuberculosis PiniB-LUX reporter strains, as measured by a robust bioluminescence signal[1].
Antitubercular agent-60 (0.125×-0.625× MIC) targets the arabinogalactan biosynthesis pathway in Mycobacterium tuberculosis, as reduced expression of embA, embB, embC, or ubiA sensitizes strains to growth inhibition[1].
Antitubercular agent-60 has reduced growth inhibition activity against Mycobacterium tuberculosis H37Rv when the embCAB operon is overexpressed, confirming targeting of the arabinogalactan biosynthesis pathway[1].
Antitubercular agent-60 (10 μM; 7 days) reduces Mycobacterium tuberculosis H37Rv burden in J774 macrophages, with activity superior to 1 μM Rifampicin (HY-B0272) after 7 days of treatment[1].
