Biological Activity
Guide (The following is our recommended solution. This solution only provides a guideline and should be modified according to your specific needs)[1].
1) Mix ATTO 740 dye with 10% sodium bicarbonate and Cetuximab antibody solution (HY-P9905) (2 mg/mL) in a certain molar ratio (such as 3 times the excess of dye) at room temperature for 2 hours.
2) Transfer the reaction solution to a Spin-X tube filled with Biogel P-6 Fine and centrifuge. The large molecule antibody-dye conjugate flows out, while the small molecule free dye is retained/removed more slowly.
3) Collect the filtrate at the bottom of the Spin-X tube (i.e., the pure antibody-dye conjugate solution), measure the final labeling degree by absorbance at 280 nm, and correct the maximum absorption wavelength of the fluorophore and each dye.
4) Measure the absorbance and perform SDS-PAGE electrophoresis to ensure no dye residue remains.
