SYTM Green

SYTM Green is a cell impermeant DNA dye that fluoresces green by binding to dsDNA (Ex/Em=503/530 nm). SYTM Green can also be used to stain bacteria, including Gram-positive and Gram-negative bacteria. Note: SYTM Green cannot penetrate living cells, and this product is equivalent to (Ex/Em=488/530 nm).

Brief Protocol for SYTM Green Nuclear Staining[1]
Cell preparation:
RAW 264.7 cells were seeded and cultured. After probe treatment, cells were fixed with 4% paraformaldehyde and washed with PBS 2-3 times.
Dual staining:
First, add Phalloidin‑TRITC to stain the cytoskeleton, incubate at room temperature for 5-10 min in the dark, then wash thoroughly with PBS. Next, add SYTM Green to stain the nucleus, incubate at room temperature for 10 min in the dark, and wash thoroughly with PBS.
Mounting and observation:
Mount slides with anti‑fluorescence quenching mounting medium. Observe under a fluorescence microscope:
SYTM Green: Ex 503 nm, Em 530 nm (green).
Key parameters:
Keep away from light throughout the procedure; for fixed cells only; sequential staining with cytoskeleton dye.
