In Vivo

PI3K-IN-64 (Compound 1) (25 mg/kg; p.o.; once daily; 4 weeks) shows potent antitumor activity in the MCF-7 breast cancer xenograft model and exhibits synergistic effects in combination with Fulvestrant (HY-13636) and Palbociclib (HY-50767)^[1].

Animal Model:

BALB/c nude mice (female, 6-8 weeks old, 18-20 g, MCF-7 xenograft model)^[1]
Dosage:

25 mg/kg (monotherapy); 25 mg/kg (combination with Fulvestrant); 25 mg/kg (triple combination with Fulvestrant and Palbociclib)
Administration:

p.o.; once daily; 4 weeks
Result:

Inhibited tumor growth in the MCF-7 xenograft model with a mean tumor volume of 148 mm³ at D28, RTV of 0.90, T/C of 29.15%, and TGI of 105.83% as monotherapy.
Inhibited tumor growth in combination with Fulvestrant with a mean tumor volume of 102 mm³, RTV of 0.60, T/C of 19.56%, and TGI of 118.89%.
Inhibited tumor growth in triple combination with Fulvestrant and Palbociclib with a mean tumor volume of 42 mm³, RTV of 0.26, T/C of 8.31%, and TGI of 136.02%.
