NCATS-SM1440
NCATS-SM1440 is a glycolysis inhibitor and metabolic regulator that targets LDHA and LDHB (IC50=0.06 μM and 0.03 μM, respectively). Upon binding to LDHA, NCATS-SM1440 blocks the glycolysis pathway and reduces lactate production. NCATS-SM1440 drives the shift of pyruvate metabolism toward mitochondrial metabolism, effectively disrupting the dependence of cancer cells on aerobic glycolysis. NCATS-SM1440 can be widely used in research related to cancer, Ewing's sarcoma, and other related conditions.

In Vitro
NCATS-SM1440 stabilizes LDHA in cells with an IC50 of 57 nM, depletes NAD+, and fully inhibits glycolysis at elevated concentrations[1].

In Vivo
NCATS-SM1440 (i.v.; p.o.) exhibits favorable pharmacokinetic properties in mice, including 73.8% oral bioavailability, measurable systemic exposure via both intravenous and oral routes, and moderate T1/2 values[2].
NCATS-SM1440 demonstrates significant in vivo antitumor activity in mouse tumor xenografts by inhibiting LDH activity, redirecting tumor cell metabolism, and synergizing with mitochondrial inhibitors[2].
