IPH5301

IPH5301 is a humanized monoclonal antibody targeting human CD73. IPH5301 potently blocks the ectonucleotidase activity of both membrane-bound and soluble CD73, preventing the degradation of AMP to immunosuppressive adenosine, thereby relieving adenosine-mediated inhibition of T cell proliferation and activation. IPH5301 shows synergistic antitumor effects with the anti-CD39 antibody IPH5201 (HY-P991444). IPH5301 can be used in research on breast cancer, melanoma, acute myeloid leukemia, and fibrosarcoma.

IPH5301 dose-dependently reverses the inhibition of T cell proliferation induced by exogenous AMP or ATP (HY-B2176) (when used in combination with an anti-CD39 antibody) in peripheral blood CD4+ and CD8+ T cells from healthy or breast cancer samples activated with anti-CD3/CD28 (HY-K0353)^[1].

IPH5301 (1-2 h) blocks the hydrolysis of AMP (HY-A0181) to adenosine by both membrane-bound and soluble CD73 in A375 human melanoma cells and MDA-MB-231 breast cancer cells (and their cell culture supernatants) in a dose-dependent and potent manner without inducing receptor internalization^[1].

IPH5301 (5-7 days) reverses ATP-mediated immunosuppression and restores T cell proliferation in a co-culture system of human monocyte-derived dendritic cells (MoDCs) and allogeneic CD4+ T cells^[1].

IPH5301 binds the CD73 dimer at a 1:1 stoichiometric ratio, trapping CD73 in an intermediate conformation incompatible with AMP hydrolysis and blocking CD73 enzymatic activity^[1].
