Cat. No.: HY-W111581 Purity: 98.5%
Handling Instructions Technical Support

Copper(II) diethyldithiocarbamate (Diethyldithiocarbamic acid copper salt) is a Copper(II) and diethyldithiocarbamate coordination polymer. Copper(II) diethyldithiocarbamate binds tightly to NPL4 and induces its aggregation, disrupting the p97-NPL4-UFD1 pathway. Copper(II) diethyldithiocarbamate causes ubiquitinated proteins accumulation and impairs waste proteins degradation, thus resulting in cell apoptosis. Copper(II) diethyldithiocarbamate inhibits tumor growth in MDA-MB-231 xenograft mice models. Copper(II) diethyldithiocarbamate can be used for the research of cancer, such as breast cancer.

Description

Copper(II) diethyldithiocarbamate (Diethyldithiocarbamic acid copper salt) is a Copper(II) and diethyldithiocarbamate coordination polymer. IC50 & Target[1]

Caspase 3

In Vitro

Copper(II) diethyldithiocarbamate (Compound CuET) (16-4000 nM; 48 h) delivered via CuET NPs and CuET@HA NPs is more cytotoxic to MDA-MB-231 breast cancer cells (IC50 115 nM and 97 nM, respectively) than to normal HEK-293 kidney cells (IC50 389 nM and 407.4 nM, respectively)[1].
Copper(II) diethyldithiocarbamate (0.1-1.0 μM; 24 h) delivered via CuET NPs induces concentration-dependent accumulation of poly-ubiquitylated proteins in MDA-MB-231 cells, confirming inhibition of the p97-NPL4 pathway[1].
Copper(II) diethyldithiocarbamate (0.5-5 μM; 24 h) delivered via CuET NPs and CuET@HA NPs induces concentration-dependent cell death in MDA-MB-231 cells after 24 h of incubation[1].
Copper(II) diethyldithiocarbamate (100-200 nM; 48 h) delivered via CuET NPs and CuET@HA NPs significantly inhibits colony formation in MDA-MB-231 cells[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Cell Cytotoxicity Assay[1]

Cell Line: MDA-MB-231 (triple negative breast cancer) cells, HEK-293 (human embryo kidney) cells
Concentration: 16, 32, 64, 125, 250, 500, 1000, 2000, 4000 nM (equivalent diethyldithiocarbamate concentration)
Incubation Time: 48 h
Result: Showed concentration-dependent cytotoxicity in MDA-MB-231 cells, with CuET@HA NPs exhibiting higher activity at each concentration.
Reached an IC50 of 115 nM in MDA-MB-231 cells (CuET NPs).
Reached an IC50 of 97 nM in MDA-MB-231 cells (CuET@HA NPs).
Reached an IC50 of 389 nM in HEK-293 cells (CuET NPs).
Reached an IC50 of 407.4 nM in HEK-293 cells (CuET@HA NPs).
In Vivo

Copper(II) diethyldithiocarbamate (Compound CuET) (1 mg/kg; i.v.) delivered via CuET NPs and CuET@HA NPs targets MDA-MB-231 xenograft tumors and shows tumor inhibitory activity in BALB/c nude mice, while maintaining low systemic toxicity[1].

MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model: BALB/c nude with MDA-MB-231 xenograft (female, 6 weeks old)[1]
Dosage: 1 mg/kg (equivalent diethyldithiocarbamate dose)
Administration: i.v.
Result: Inhibited tumor growth significantly over 14 days, with CuET@HA NPs showing the most potent efficacy (slowest tumor growth rate and smallest final tumor size).
Induced the most distinct nuclear necrosis in tumor tissue on H&E staining (CuET@HA NP-treated mice).
Increased caspase-3 activation to the highest level on immunofluorescence staining (CuET@HA NP-treated mice).
Caused the greatest extent of apoptosis on TUNEL staining (CuET@HA NP-treated mice).
Caused no noticeable mouse weight loss or pathological changes in major organs (heart, liver, spleen, lung, kidney) during treatment.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
