DHEA Benzophenone-biotin is a photoactive DHEA (HY-14650) analog containing Biotin (HY-B0511) and Benzophenone (HY-Y0546) groups. DHEA Benzophenone-biotin inhibits plasma membrane binding of DHEA, activates endothelial NO synthase. DHEA Benzophenone-biotin cross-links to plasma membrane DHEA binding sites upon UV irradiation to identify associated proteins.

Biological Activity
DHEA Benzophenone-biotin (Compound DHEA–BP–Bt) (30 min) binds with high affinity to the plasma membrane DHEA (HY-14650) binding site on BAEC, with a Kd of 3.49 nM[1].
DHEA Benzophenone-biotin (10 μM; 1 h) forms a complex with avidin-agarose that retains high affinity for the plasma membrane DHEA binding site on BAEC[1].
DHEA Benzophenone-biotin (2 nM; 30 min) binds specifically to BAEC plasma membranes in a DHEA-inhibitable manner, and can simultaneously bind its plasma membrane site and streptavidin-FITC[1].
DHEA Benzophenone-biotin (5 nM; 1 h) irreversibly cross-links to the plasma membrane DHEA binding site on BAEC with 62% efficiency after 15 min of 365 nm UV irradiation, blocking subsequent [3H]HDHEA binding[1].
DHEA Benzophenone-biotin (10 pM-100 nM; 15 min) activates eNOS in BAEC in a concentration-dependent manner, reaching a maximal 81.6% increase in activity at 10 nM, retaining the biological activity of unmodified DHEA[1].
DHEA Benzophenone-biotin (1 nM) specifically cross-links to BAEC plasma membrane proteins of approximately 55 kDa (detected by both live cell labeling and ligand blot) and 80 kDa (detected by live cell labeling only) in a DHEA-inhibitable manner[1].
