DMG-PEG3400-LyP-1

DMG-PEG3400-LyP-1 is specifically designed for active targeting nanomedicine applications. It can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against p32/gC1qR protein on the surface of lymphatic vessels, tumor-associated macrophages, and tumor cells. Main applications include: lymphatic metastasis targeting, tumor microenvironment modulation, and tumor imaging.
Composition and role of each component:
1. DMG (dimyristoylglycerol): Lipid anchoring group-a diacylglycerol with two C14 chains, providing moderate membrane anchorage; its short-chain, high-fluidity characteristics enable faster dissociation of PEG-lipids from nanoparticle surfaces, facilitating endosomal escape and cellular uptake.
2. PEG (polyethylene glycol): Hydrophilic polymer spacer arm that forms a steric corona around nanoparticles, reducing opsonization and reticuloendothelial system (RES) clearance, prolonging circulation half-life, and providing spatial separation between the lipid surface and targeting ligand.
3. LyP-1 (LyP-1 peptide (CGNKRTRGC)): Targeting ligand-a cyclic 9-amino acid peptide that specifically binds to p32/gC1qR protein, which is highly expressed on tumor-associated macrophages, lymphatic endothelial cells, and certain tumor cells.
