DNP Phosphoramidite
DNP Phosphoramidite is a non-nucleoside-based phosphoramidite carrying a 2,4-dinitrophenyl (DNP) hapten group, which enables multiple labeling of oligonucleotides during solid-phase synthesis. DNP Phosphoramidite binds to anti-DNP antibodies, allowing the detection of labeled oligonucleotides via enzyme-linked colorimetric techniques.

Biological Activity
DNP Phosphoramidite (0.15 M; 5 h deprotection, 30 mins detritylation) enables efficient solid-phase labelling of oligonucleotides, with "trityl-on" synthesis preventing degradation during 5-hour 60°C concentrated ammonia deprotection, and resulting labelled oligonucleotides showing increased lipophilicity for easy HPLC purification[1].
DNP Phosphoramidite enables DNP-labelled 24-mer oligonucleotides to exhibit optimal colorimetric dot blot detection sensitivity with 3 DNP labels, achieving limits comparable to biotin-labelled oligonucleotides regardless of spacer arm length[1].
