Biological Activity
Guide (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
1. Solution preparation[3]
1.1 Preparation of stock solution
Solvent: DMSO
Concentration: 10 mM.
1.2 Preparation of working solution
Dilute to 1-10 μM with PBS or cell culture medium (optimized according to the experiment).
Note: The working solution should be prepared and used immediately. Keep it away from light.
2. Cell staining
2.1 Cells are plated in clear-bottomed black 96-well microplates.
2.2 Add Monochlorobimane working solution and incubate at 37°C for 30 minutes (optimized according to the experiment).
2.3 The fluorescence intensity is measured with fluorescence microscopy or Fluorescence microplate reader (Ex/Em = 380/480 nm).

Protocol
Monochlorobimane (mCB) is added to the second half liver tissue homogenate to a final concentration of 100 mM along with glutathione S-transferase (1 U/mL) obtained from equine liver; the homogenate is then allowed to incubate at room temperature for 30 min. The Glutathione (GSH)-Monochlorobimane adduct is measured in a microtiter reader[1].
