HBsAg-IN-1
HBsAg-IN-1 is an orally active, highly liver-selective inhibitor of HBsAg production, with an EC50 of 0.63 nM. HBsAg-IN-1 shows no neurite growth inhibitory activity. HBsAg-IN-1 can be used in studies related to hepatitis B virus (HBV) infection.

Biological Activity
HBsAg-IN-1 (Compound (6S,10S)-57) (12.8-40000 nM) potently inhibits HBsAg production in HepG2.2.15 cells, with an EC50 of 0.63 nM, and exhibits extremely low cytotoxicity in these cells (CC50 >200 nM)[1].
HBsAg-IN-1 (12.5-100 μM; 24 h) exhibits negligible cytotoxicity in SH-SY5Y cells, with a CC50 >100 μM[1].
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Cell Line:HepG2.2.15
-
Concentration:12.8-40000 nM
-
Incubation Time:6 days
-
Result:Potently inhibited HBsAg production with an EC50 of 0.63 nM.
Exhibited CC50 values >200 nM, indicating negligible cytotoxicity at the tested concentrations.
-
Cell Line:SH-SY5Y
-
Concentration:12.5-100 μM
-
Incubation Time:24 h
-
Result:Showed negligible cytotoxicity, with a CC50 >100 μM (no ≥50% reduction in cell viability at the highest tested concentration of 100 μM).
MedChemExpress (MCE) has not independently confirmed the accuracy of these methods. They are for reference only.

Animal Model:C57BL/6 (male, inoculated with AAV vector carrying 1.3-fold overlength genotype D HBV genome, dosing initiated 4 weeks post-inoculation)[1]
-
Dosage:8 mg/kg; 24 mg/kg
-
Administration:p.o.; once daily; 15 days
-
Result:Reduced serum HBsAg by 0.78 log10 and serum HBeAg by 0.31 log10 relative to predose baseline at day 15 with 8 mg/kg dose.
Reduced serum HBsAg by 0.97 log10 and serum HBeAg by 0.41 log10 relative to predose baseline at day 15 with 24 mg/kg dose.
Caused no significant body weight changes at either dose.
Induced significant reduction in liver weight and liver coefficient in the 24 mg/kg group.
Maintained serum creatinine and ALT levels within normal ranges across both dose groups.
Resulted in lower antigen rebound compared to the RG7834 control group following treatment withdrawal.
