Assemblin (herpesvirus protease) is a herpes-virus serine protease in family S21 involved in breakdown of the scaffold protein during late virion assembly, inhibited by diisopropyl fluorophosphate, and the type example of family S21 with catalytic residues His-Ser-His (a combination unknown in any other peptidase) and a unique protein fold; known from herpesviruses including cytomegalovirus, Epstein-Barr virus and human herpesvirus 3; it cleaves Ala-Ser and Ala-Ala bonds in the scaffold protein. By processing the scaffold it enables herpes capsid maturation.
