DOTA-NHS-ester TFA is a bifunctional reagent containing a DOTA chelating group and an NHS ester. DOTA-NHS-ester TFA can be conjugated to proteins/antibodies/HSA via primary amino groups for radiolabeling with radiometals, and when combined with 16-amino-1-hexadecanethiol hydrochloride, it enables the construction of self-assembled monolayers on gold surfaces for lectin microarrays. DOTA-NHS-ester TFA can be used to prepare DOTA-HSA-Z (HER2:342) imaging probes, as well as to construct Trop2-targeted radiotherapeutic and diagnostic probes for pancreatic cancer.

DOTA-NHS ester (3.78 mg; 2 h) TFA successfully conjugates to HSA, yielding a DOTA-HSA bioconjugate with approximately two DOTA molecules per HSA molecule[1].
DOTA-NHS-ester (10 mM; overnight) TFA in combination with 16-amino-1-hexadecanethiol hydrochloride successfully modifies gold chip surfaces, producing a rough surface topography and characteristic infrared absorption peaks consistent with covalent coupling of the reagent[2].
DOTA-NHS-ester (5-20 mM; 2 h) TFA modification concentration on gold biochips positively correlates with IgG protein binding capacity, with 10 mM selected as the optimal working concentration balancing performance and reagent efficiency[2].
DOTA-NHS-ester (10 mM; overnight) TFA modification of gold biochips proceeds effectively at both 25 °C and 37 °C, with 25 °C selected as the optimal working temperature for practical convenience[2].
DOTA-NHS-ester (10 mM; overnight) TFA modified gold biochips support specific, concentration-dependent binding of ConA lectin and its corresponding antibody, validating the platform's suitability for lectin-based assays[2].

PSP–NH2 was incubated with DOTA–NHS–ester for 1 h and conjugated with mPEG–NHS. Then, the particles were mixed with 64Cu (OAc)2 at 50 ℃ for 1 h to obtain the 64Cu-labelled PSP. PSP@Ab@PEG exhibited increased blood circulation in BALB/c mice with blood clearance half-life periods found to be 12 min.
